Cell Injection Procedure

Cell injection procedure is a technique for delivering substances directly into individual cells, enabling controlled manipulation of cellular contents and function. In a typical microinjection, a fine glass micropipette penetrates the cell membrane and deposits a measured volume of DNA, RNA, proteins, drugs, or other materials into the cytoplasm or nucleus. Researchers use this approach to study gene expression, cell signaling, development, and responses to treatment while observing effects in selected cells. Although technically demanding, cell injection provides precise control over dose, location, and timing, supporting experiments in molecular biology, developmental biology, and biomedical research.

This video demonstrates the procedure of intravenously injecting cancer cells into the chicken chorioallantoic membrane vasculature to generate a CAM cancer metastasis model. The injected cells eventually form scattered single-cancer cell colonies, allowing for the study of cancer invasion and metastasis.

We describe an intracisternal injection that employs a needle bent at the tip that can be stabilized to the skull, thus eliminating the risk of damage to the underlying parenchyma. The approach can be used for genetic fate mapping and manipulations of leptomeningeal cells and for tracking cerebrospinal fluid movement.

Here we present a community accepted protocol in multimedia format for subretinally injecting a bolus of RPE cells in rats and mice. This approach can be used for determining rescue potentials, safety profiles, and survival capacities of grafted RPE cells upon implantation in animal models of retinal degeneration.
