Collagen Gel Contraction

Collagen gel contraction is a biological process and laboratory assay that measures how cells compact a three-dimensional collagen matrix, providing insight into tissue remodeling and mechanical force generation. In a hydrated collagen lattice, embedded cells such as fibroblasts attach to collagen fibers and use actomyosin-generated contractile forces to pull the network inward, reducing gel area or volume over time. The extent and rate of contraction depend on cell type, matrix composition, cell density, and culture conditions. In biology, this model helps investigate wound healing, fibrosis, cell-matrix interactions, and the effects of drugs or genetic changes on connective-tissue remodeling.

Collagen is a core component of the ECM, and provides essential cues for several cellular processes ranging from migration to differentiation and proliferation. Provided here is a protocol for embedding cells within 3D collagen hydrogels, and a more advanced technique for generating randomized or aligned collagen matrices using PDMS microchannels.

In this work, we present a technique for the rapid fabrication of living vascular tissues by direct culturing of collagen, smooth muscle cells and endothelial cells. In addition, a new protocol for the mechanical characterization of engineered vascular tissues is described.

Dissection and Culture of Chick Statoacoustic Ganglion and Spinal Cord Explants in Collagen Gels for Neurite Outgrowth Assays

We demonstrate how to dissect and culture chick E4 statoacoustic ganglion and E6 spinal cord explants. Explants are cultured under serum-free conditions in 3D collagen gels for 24 hours. Neurite responsiveness is tested with growth factor-supplemented medium and with protein-coated beads.
