Diabetes Complications

Diabetes complications are health problems that develop when diabetes disrupts normal physiology, making them important outcomes to understand in biology and clinical research. Persistent hyperglycemia can damage blood vessels and nerves through oxidative stress, inflammation, and the accumulation of advanced glycation end products, leading to microvascular and macrovascular injury. These processes contribute to conditions such as retinopathy, nephropathy, neuropathy, cardiovascular disease, and impaired wound healing. Studying diabetes complications helps researchers identify risk factors, develop strategies for glucose management and early detection, and evaluate treatments that reduce long-term tissue damage and improve patient outcomes.

An Ex Vivo Tissue Culture Model for Fibrovascular Complications in Proliferative Diabetic Retinopathy

Here, we present a protocol to study the pathophysiology of proliferative diabetic retinopathy by using patient-derived, surgically-excised, fibrovascular tissues for three-dimensional native tissue characterization and ex vivo culture. This ex vivo culture model is also amenable for testing or developing new treatments.

Metabolic memory is the phenomenon by which diabetic complications persist and progress unimpeded even after euglycemia is achieved pharmaceutically. Here we describe a diabetes mellitus zebrafish model which is unique in that it allows for the examination of the mitotically transmissible epigenetic components of metabolic memory in vivo.

Transplantation of Pancreatic Islets Into the Kidney Capsule of Diabetic Mice

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.
