Ex Vivo Cornea Model

An ex vivo cornea model is a laboratory system that maintains excised corneal tissue outside the body to study ocular biology under controlled conditions. It works by placing the cornea in an organ-culture environment that supplies nutrients and regulates temperature, hydration, and gas exchange, helping preserve its layered structure and cellular responses. These models support investigation of epithelial wound healing, corneal infection, drug penetration, toxicity, and surgical treatments, providing a biologically relevant bridge between simplified cell cultures and in vivo studies. In biology and ophthalmic research, they improve understanding of tissue behavior while enabling more controlled evaluation of experimental interventions.

The cornea is unique in that it lacks vascular tissues. However, robust blood vessel growth and survival can be induced in the cornea by potent angiogenic factors. Therefore, the cornea can provide with us a valuable tool for angiogenic studies. This protocol demonstrates how to perform the mouse model of cornea pocket assay and how to assess the angiogenesis induced by angiogenic factors using this model.
