Fc Gamma Receptor

Fc gamma receptors (FcγRs) are cell-surface immune receptors that bind the Fc region of immunoglobulin G (IgG), linking antibody recognition to cellular responses. When IgG coats a target, receptor binding clusters FcγRs and triggers signaling through immunoreceptor tyrosine-based activation or inhibition motifs, depending on the receptor, leading to phagocytosis, cytokine production, antibody-dependent cellular cytotoxicity, or reduced activation. Expressed on macrophages, neutrophils, natural killer cells, and dendritic cells, FcγRs regulate host defense, inflammation, and immune-complex clearance. Their biology is important in infection research, autoimmune disease, cancer immunology, and the design of therapeutic antibodies.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.
