Claudin-7 Knockout

Claudin-7 knockout is a genetic approach that eliminates expression of the CLDN7 gene, which encodes a tight-junction protein involved in epithelial barrier organization and cell-cell adhesion. In cancer research, researchers disrupt CLDN7 in cultured cells or animal models, often by introducing targeted mutations that prevent production of functional claudin-7, then compare the resulting cells with controls. This model helps reveal how loss of epithelial integrity affects cell polarity, migration, invasion, and interactions with the tumor microenvironment. Claudin-7 knockout systems can therefore clarify the protein’s role in cancer progression and support evaluation of pathways or treatments associated with epithelial stability and metastasis.

This video describes an in vitro Caspase-3/7 luminescent assay to quantify cell death or apoptosis. The assay measures luminescence produced following caspase-3/7 cleavage of a pro-luminescent DEVD substrate. The luminescence generated is proportional to caspase-3/7 activity.

Production of Apolipoprotein C-III Knockout Rabbits using Zinc Finger Nucleases

Recent development in gene targeting tools makes production of knockout (KO) rabbits possible. In the present work, we generated five Apolipoprotein (Apo) C-III KO rabbits using Zinc Finger Nucleases (ZFN). This work demonstrated that ZFN is a highly efficient method to produce KO rabbits.
