Gut Mucosal Model

A gut mucosal model is an experimental system that reproduces key features of the intestinal lining, including its epithelial barrier, mucus layer, and interactions with surrounding cells, to study gastrointestinal biology and disease. These models typically organize intestinal epithelial cells into a polarized, selectively permeable surface, while mucus-producing cells and supporting stromal or immune components help recreate the local microenvironment. In cancer research, gut mucosal models enable controlled investigation of tumor initiation, invasion, barrier disruption, inflammation, and responses to anticancer treatments. They can complement animal studies by providing human-relevant platforms for analyzing tumor–tissue interactions and evaluating therapeutic strategies.

This manuscript describes the production, characterization and potential uses of a tissue engineered 3D esophageal construct prepared from normal primary human esophageal fibroblast and squamous epithelial cells seeded within a de-cellularized porcine scaffold. The results demonstrate the formation of a mature stratified epithelium similar to the normal human esophagus.

A mouse model of human endoscopic skull base reconstruction has been developed that creates a semipermeable interface between the brain and nose using nasal mucosal grafts. This method allows researchers to study delivery to the central nervous system of high molecular weight therapeutics which are otherwise excluded by the blood-brain barrier when administered systemically.
