Myc-cap Matrigel

Myc-CaP Matrigel refers to embedding Myc-CaP prostate cancer cells in Matrigel, a basement membrane-derived extracellular matrix, to create a three-dimensional model of tumor growth. The gel provides structural support and extracellular matrix signals that promote cell attachment, proliferation, and organization under controlled culture conditions, more closely resembling aspects of the tumor microenvironment than conventional two-dimensional culture. This system supports studies of prostate cancer biology, tumor-cell behavior, therapeutic responses, and interactions between cancer cells and their surrounding matrix. It can also help researchers evaluate experimental treatments and generate reproducible in vitro evidence before advancing to animal or clinical studies.

Here, we describe a technique to isolate the side population cells from a zebrafish model of myc-induced T-cell acute lymphoblastic leukemia (T-ALL). This side population assay is highly sensitive and is described for zebrafish T-ALL, but it may be applicable to other malignant and non-malignant zebrafish cell types.
