Sirna Functional Screening

siRNA functional screening is a gene-silencing method that systematically tests how reducing individual gene expression affects a cellular phenotype, helping researchers identify genes that control cancer behavior. Researchers introduce libraries of small interfering RNAs (siRNAs) into cancer cells; each siRNA is loaded into the RNA-induced silencing complex, which recognizes complementary messenger RNA and promotes its degradation or suppresses its translation. By measuring outcomes such as cell growth, viability, apoptosis, invasion, or drug sensitivity, scientists can connect gene function with pathways that support tumor development and treatment resistance. In cancer research, these screens reveal genetic dependencies and potential therapeutic targets, while follow-up experiments help confirm whether candidate genes can guide biomarker development or precision treatment strategies.

Here, we describe a methodology to perform a targeted siRNA “ubiquitome” screen to identify novel ubiquitin and ubiquitin-like regulators of the HIF1A-mediated cellular response to hypoxia. This can be adapted to any biological pathway where a robust read out of reporter activity is available.
