Tryptophan Metabolism Imaging

Tryptophan metabolism imaging is the use of labeled molecular tracers and imaging technologies to visualize how cells and tissues process tryptophan, an amino acid that feeds several biochemical pathways. In positron emission tomography, radiolabeled tryptophan analogs emit positrons that produce detectable signals, allowing researchers to estimate tracer uptake, transport, and conversion into metabolites associated with serotonin synthesis or the kynurenine pathway. This chemistry-based approach links molecular reactions to spatial information in living systems. It supports studies of brain function, immune activation, cancer metabolism, and neurological disease by revealing pathway activity that may change before structural abnormalities become apparent.

Chemical Modification of the Tryptophan Residue in a Recombinant Ca2+-ATPase N-domain for Studying Tryptophan-ANS FRET

ANS binds to the Ca2+-ATPase recombinant N-domain. Fluorescence spectra display a FRET-like pattern upon excitation at a wavelength of 295 nm. NBS-mediated chemical modification of Trp quenches the fluorescence of the N-domain, which leads to the absence of energy transfer (FRET) between the Trp residue and ANS.

Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.
