Multiphoton Excitation

Multiphoton excitation is a nonlinear optical process in which a molecule absorbs two or more photons nearly simultaneously to reach an excited electronic state, enabling fluorescence or other optical signals. It typically uses pulsed near-infrared light, and excitation occurs most strongly at the microscope focal point because the required photon coincidence is unlikely elsewhere. In developmental biology, this approach supports three-dimensional imaging of living embryos, organoids, and tissues while limiting out-of-focus illumination and associated photodamage. By capturing cell movements, divisions, and tissue remodeling over time, multiphoton microscopy helps connect dynamic cellular behavior with the mechanisms that shape developing organisms.

Multiphoton Microscopy of Cleared Mouse Brain Expressing YFP

Multiphoton microscopy of whole mouse organs is possible by optically clearing the organ before imaging, but not all protocols preserve the fluorescent signal of fluorescent proteins. Using an optical clearing method with ethanol-based dehydration and benzyl alcohol:benzyl benzoate clearing, we show high-resolution multiphoton images of whole mouse brain expressing YFP.

Culture of Adult Transgenic Zebrafish Retinal Explants for Live-cell Imaging by Multiphoton Microscopy

Zebrafish retinal regeneration has mostly been studied using fixed retinas. However, dynamic processes such as interkinetic nuclear migration occur during the regenerative response and require live-cell imaging to investigate the underlying mechanisms. Here, we describe culture and imaging conditions to monitor Interkinetic Nuclear Migration (INM) in real-time using multiphoton microscopy.

Nerve Excitability Assessment in Chemotherapy-induced Neurotoxicity

This abstract describes a novel method to assess the development of neurotoxicity in patients receiving chemotherapy treatment. While conventional assessment methods are limited in their ability to detect early changes in nerve function, nerve excitability techniques provide early identification of patients at risk of severe neurotoxicity and insight into pathophysiology.
