Progeny Testing Strategy

Progeny testing strategy is a genetic evaluation method that estimates an individual’s heritable contribution by examining traits in its offspring. The approach involves mating a candidate parent with selected partners, then measuring offspring phenotypes or genotypes across families and, when possible, under consistent conditions to separate inherited effects from environmental variation. In developmental biology, progeny testing helps link parental alleles to embryonic development, growth, morphology, fertility, and other inherited traits. It can reveal recessive mutations, assess genetic variation, and guide the selection of model organisms or breeding lines for studies of development and disease.

Shifting Zebrafish Lethal Skeletal Mutant Penetrance by Progeny Testing

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.

Barnes Maze Testing Strategies with Small and Large Rodent Models

The dry-land Barnes maze is widely used to measure spatial navigation ability in response to mildly aversive stimuli. Over consecutive days, performance (e.g. latency to locate escape cage) of control subjects improves, indicative of normal learning and memory. Differences between rats and mice necessitate apparatus and methodology changes that are detailed here.

While infiltrating macrophages are continuously recruited to adult tissues from circulating precursors, resident macrophages seed their tissue during development, where they are maintained without further input from progenitors. The progenitors for resident macrophages were recently identified. Here, we present methods for the genetic fate mapping of the resident macrophage progenitors.

Set-shifting, a form of behavioral flexibility, requires an attentional shift from one stimulus dimension to another. We extended an established rodent set-shifting task1 by requiring attention to different stimuli according to context. The task was combined with specific lesions to identify neuron subtypes underlying a successful shift.
