Total Lipid Determination

Total lipid determination is the analytical measurement of the combined lipid content in a biological, sediment, water, or other environmental sample, providing information about organic matter and biological material. In a typical solvent-extraction approach, lipids are separated from nonlipid components according to their solubility, the extract is collected, and solvent removal permits gravimetric or instrumental quantification. The resulting measurement supports assessment of microbial and aquatic biomass, sediment and wastewater composition, and lipid-associated contaminant partitioning. Comparing total lipid levels across locations or time points can help evaluate ecosystem conditions, environmental change, and the performance of treatment or remediation processes.

This protocol is for the determination of lipids in seawater and biological specimens. Lipids in filtrates are extracted with chloroform or mixtures of chloroform and methanol in the case of solids. Lipid classes are measured by rod thin-layer chromatography with flame ionization detection and their sum gives the total lipid content.

We introduce a fast fluorescence-based assay that monitors the rate of fluorescence quenching as a measure of gramicidin channel activity. The gramicidin channels are used as molecular force transducers to monitor changes in lipid bilayer properties as sensed by bilayer spanning proteins.

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.
