Cell Lysis Fragmentation

Cell lysis and fragmentation is the controlled disruption of cells to release genomic DNA and other intracellular components for genetic analysis. Lysis breaks the plasma membrane, while mechanical forces, detergents, enzymes, or chemical conditions can disrupt cell walls and proteins; subsequent handling fragments or shears released DNA to produce material suited to a specific assay. In genetics, this preparation supports DNA extraction, library construction, PCR, sequencing, and other workflows, but excessive disruption can reduce DNA integrity and insufficient lysis can lower yield. Controlling the method and conditions therefore affects sample quality, reproducibility, and the reliability of downstream genomic measurements.

Tissue Homogenization and Cell Lysis

2023

Tissue homogenization involves disintegrating tissue architecture and lysing cells, and is an early step in isolating and analyzing cellular components. The method used for homogenization depends on the sample type, the amount of sample available, the analyte to be obtained, and the sensitivity of the method. These methods are broadly classified as mechanical and non-mechanical methods. Mechanical methods of tissue homogenization These methods rely on applying external physical force to disrupt...

Source: Wunderlin, T., et al. Physical Isolation of Endospores from Environmental Samples by Targeted Lysis of Vegetative Cells. J. Vis. Exp. (2016)This video demonstrates a method to isolate endospores from an environmental sample containing both vegetative cells and various bacteria. The procedure involves filtration, mechanical disruption, and selective chemical treatment to weaken and lyse vegetative cells while preserving endospores. The final step isolates the resilient endospores through...

Habitat Fragmentation

2020

Habitat fragmentation describes the division of a more extensive, continuous habitat into smaller, discontinuous areas. Human activities such as land conversion, as well as slower geological processes leading to changes in the physical environment, are the two leading causes of habitat fragmentation. The fragmentation process typically follows the same steps: perforation, dissection, fragmentation, shrinkage, and attrition. Perforation and dissection often occur during the initial stages of...
