Large-scale Culture Plate

A large-scale culture plate is a laboratory platform that supports the growth and comparison of many biological samples in parallel, making it valuable for genetics and other fields that require reproducible, higher-throughput experiments. Its wells keep cultures physically separated while providing defined nutrient media and controlled incubation conditions, allowing researchers to inoculate, replicate, monitor, and compare samples under consistent conditions. In genetic studies, these plates can support clonal growth, phenotype screening, selection experiments, and genotype–phenotype analysis. By increasing sample capacity while reducing handling variability, large-scale culture plates help researchers evaluate genetic effects more efficiently and generate data suitable for systematic analysis.

To use Caenorhabditis elegans (C. elegans) in omics research, a method is needed to generate large populations of worms where a single sample can be measured across platforms for comparative analyses. Here, a method to culture C. elegans populations on large-scale culture plates (LSCPs) and to document population growth is presented.

The protocols described allow laboratories to perform scalable, adherent stem cell culture in high throughput with minimal labor, experience and equipment investment cost using a programmable liquid handling robot and 96-well plates. iPSCs passaged more than 20 times on this system maintained pluripotency, normal karyotypes and differentiated into cardiomyocytes.

The floor plate is a crucial structure of the developing spinal cord, providing diffusible signals to progenitors and differentiating neurons. We describe a method to produce floor-plate conditioned medium, to apply it to fresh spinal cord tissue, and to assess the consequences on proteins of interest by biochemistry.
