Targeted Dna Insertion

Targeted DNA insertion is a genetic engineering method that places a chosen DNA sequence at a defined location in a genome, enabling controlled changes to gene function or regulation. The process typically uses a programmable nuclease, such as CRISPR-associated enzymes, to create a break at the target site, while a donor DNA template guides repair through homology-directed recombination or another insertion pathway. In genetics research, this approach supports precise gene tagging, correction of disease-associated variants, transgene integration, and functional studies of regulatory elements. Compared with random insertion, targeted DNA insertion improves control over genomic position and helps researchers interpret resulting phenotypes.

Bisulfite amplicon sequencing (BSAS) is a method for quantifying cytosine methylation in targeted genomic regions of interest. This method uses bisulfite conversion paired with PCR amplification of target regions prior to next-generation sequencing to produce absolute quantitation of DNA methylation at a base-specific level.

We present a method of targeted ancient DNA sequence retrieval, which we used to reconstruct the complete mitochondrial genomes of five Neandertal individuals. Comparison of these sequences with present day humans suggests that Neandertals had a long term low effective population size.
