Cellular Compartment Purification

Cellular compartment purification is a group of methods used to isolate defined regions of a cell, such as the nucleus, cytoplasm, membranes, or organelles, for focused biochemical and functional analysis. The process typically combines controlled cell disruption with differential centrifugation or density-gradient separation, in which cellular components are distinguished by size, density, and sedimentation behavior while preserving relevant molecular features. In immunology and infection research, purified compartments help researchers determine where host immune signaling, pathogen replication, or microbial effector proteins occur within infected cells. These preparations support protein and nucleic acid analysis, microscopy validation, and studies of compartment-specific mechanisms that shape cellular responses to infection.

We describe a protocol allowing the purification of the mouse brain's vascular compartment. Isolated brain vessels include endothelial cells linked by tight junctions and surrounded by a continuous basal lamina, pericytes, vascular smooth muscle cells, as well as perivascular astroglial membranes.

Two techniques for isolating cellular lipid droplets from 1) yeast cells and 2) human placentas are presented. The centerpiece of both procedures is density gradient centrifugation, where the resulting floating layer containing the droplets can be readily visualized by eye, extracted, and quantified by Western Blot analysis for purity.
