Magnaporthe Grisea

Magnaporthe grisea is a filamentous ascomycete fungus best known as the causal agent of rice blast, a major disease that threatens cereal production worldwide. Infection begins when a conidium germinates on the plant surface and differentiates into a melanized appressorium, which generates concentrated turgor pressure to drive a penetration peg through the cuticle. After entering host tissue, the fungus grows biotrophically and deploys secreted effectors that alter plant immune signaling before lesions develop. Studying this pathogen clarifies fungal virulence, pattern-triggered immunity, and host-pathogen coevolution, while supporting resistant crop breeding, disease surveillance, and targeted disease control.

This is a straightforward protocol of a barley leaf sheath assay using minimal reagents and common laboratory equipment (including a basic smartphone). The purpose is to visualize the early infection process of blast disease in labs without access to advanced microscopy equipment.

Establishment of Rice Plant Leaf Infection through Fungal Inoculation via Wounding

This video demonstrates the establishment of rice plant leaf infection through wounding inoculation with Magnaporthe grisea mycelium. The fungal spore-derived germ tube penetrates the epidermal layer, resulting in tissue infection and lesions on the leaves. This method aids pathotype screening and improves plant resistance understanding for molecular breeding.

A Spray Inoculation Technique to Establish Fungal Infection in Rice Plant Leaves
