Posterior Semicircular Canal

The posterior semicircular canal is one of three fluid-filled canals in the inner ear that detects rotational head movement and contributes to balance. When the head rotates, endolymph lags behind because of inertia, deflecting the canal’s cupula and bending sensory hair cells in the ampulla; this changes vestibular nerve signaling to the brain, which coordinates eye movements and posture. In clinical and research settings, understanding this pathway supports evaluation of vertigo, including benign paroxysmal positional vertigo caused by displaced otoconia, and helps distinguish mechanical vestibular dysfunction from inflammation or infection affecting the inner ear. Its anatomy also provides a framework for studying how immune-mediated injury can disrupt balance.

In this study, we describe the posterior semicircular canal approach as a reliable method for inner ear gene delivery in neonatal mice. We show that gene delivery through the posterior semicircular canal is able to perfuse the entire inner ear.
