Protein Co-localization Analysis

Protein co-localization analysis is a microscopy-based technique used to determine whether two or more proteins occupy the same cellular or tissue region, providing insight into their potential functional relationship. The method typically labels target proteins with distinct fluorescent tags or antibodies, acquires images under controlled conditions, and compares overlapping signal patterns using spatial measurements such as correlation or overlap coefficients. In immunology and infection research, it can reveal receptor clustering, protein recruitment to immune synapses, pathogen entry sites, or interactions between host and microbial proteins. These measurements help connect protein distribution with signaling, cellular defense, infection mechanisms, and treatment responses.

Plant intercellular connections, the plasmodesmata (Pd), play central roles in plant physiology and plant-virus interactions. Critical to Pd transport are sorting signals that direct proteins to Pd. However, our knowledge about these sequences is still in its infancy. We describe a strategy to identify Pd localization signals in Pd-targeted proteins.

This manuscript provides a detailed protocol for the study of protein-protein interactions (PPIs) and co-localization using murine mammary glands. These techniques allow for the investigation of the dynamics of the physical association between intercellular junctions at different developmental stages.

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.

This video demonstrates a method for monitoring infected cell protein 0, ICP0 trafficking in herpes simplex virus-1 infection. Post-de novo synthesis, ICP0 translocates to the nucleus, later moving to the cytoplasm during infection progression. Immunofluorescence microscopy reveals and analyzes the protein's subcellular localization, offering insights into its trafficking across infection phases.
