T-cell Depletion

T-cell depletion is the reduction or removal of T lymphocytes from blood, tissues, or a biological sample, a process that can profoundly alter adaptive immune responses. It occurs through mechanisms such as antibody-mediated cell killing, complement activation, targeted drug treatment, or infection-driven loss of T cells, depending on the experimental or clinical context. In immunology and infection research, controlled depletion helps define how T cells contribute to pathogen clearance, immune regulation, and immunopathology. It is also used in transplantation and cellular therapies to reduce graft rejection or graft-versus-host disease, while monitoring depletion informs studies of immune recovery and susceptibility to infection.

Human tumor xenografts are vascularized and infiltrated by cells of mouse origin during the growth phase in vivo. To circumvent the bias caused by these contaminating cells during downstream analysis, we developed a method allowing for comprehensive depletion of all mouse cells by magnetic cell sorting.

Here, we present the real-time monitoring of cell migration in a wound-healing assay using TIP60-depleted MCF10A breast epithelial cells. The implementation of live-cell imaging techniques in our protocol allows us to analyze and visualize single-cell movement in real time and across time.

Depletion and Reconstitution of Macrophages in Mice

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.
