Fetal Cardiac Function

Fetal cardiac function describes how the developing heart generates blood flow and maintains circulation before birth, providing essential information about fetal growth and well-being. Because the lungs are not yet used for oxygen exchange, oxygenated blood from the placenta is distributed through specialized pathways, including the foramen ovale and ductus arteriosus, while coordinated myocardial contraction supports systemic and placental circulation. Clinicians assess these processes with fetal echocardiography, ultrasound, and Doppler measurements of heart structure, rhythm, blood flow, and ventricular performance. Evaluation can identify congenital heart defects, arrhythmias, and signs of hemodynamic compromise, guiding monitoring and perinatal care.

Fetal and perinatal death is a common feature when studying genetic alterations affecting cardiac development. High-frequency ultrasound imaging has improved 2-D resolution and can provide excellent information on early cardiac development and is an ideal method to detect the impact on cardiac structure and function prior to death.

We describe examination of fetal cardiac function with contemporary functional fetal echocardiography and fetoplacental Doppler ultrasound using the VisualSonics VEVO 2100 microultrasound in a surgically induced model of intrauterine fetal growth restriction in a rabbit.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Langendorff-mode isolated heart perfusion, in conjunction with 31P NMR spectroscopy, combines the fields of biochemistry and physiology into one experiment. The protocol allows for the dynamic measurement of high energy phosphate content and turnover in the heart while concurrently monitoring physiologic function. When performed correctly, this is a valuable technique in the assessment of cardiac energetics.
