Genetic Mouse Models

Genetic mouse models are laboratory mice whose DNA has been altered to reproduce specific human genes, mutations, or disease-related traits, making them valuable tools in medical research. Researchers create transgenic, knockout, knock-in, or conditional alleles through targeted genome modification, often controlling gene activity in particular tissues or at selected times with systems such as Cre-lox recombination. These models help clarify disease mechanisms, reveal gene function, and measure how pathological changes develop in living organisms. They also support preclinical evaluation of drugs, gene therapies, and other interventions, although differences between mouse and human biology require careful interpretation when translating findings to patient care.

Transgenesis, or the use of genetic engineering to alter gene expression, is widely used in the field of developmental biology. Scientists use a number of approaches to alter the function of genes to understand their roles in developmental processes. This includes replacement of a gene with a nonfunctional copy, or adding a visualizable tag to a gene that allows the resultant fusion protein to be tracked throughout development. In this video, the viewers will learn about the principles behind...

(2016)This video demonstrates stable genetic modification of neural stem cells (NSCs) in the ventricular-subventricular zone (V-SVZ) through stereotaxic injection of a virus-carrying therapeutic gene in a mouse model. The virus enters NSCs, undergoes reverse transcription, integrates into the genome, and...
