Pegylated Nanoparticles

Pegylated nanoparticles are nanoscale drug-delivery systems coated or chemically linked with polyethylene glycol (PEG), a hydrophilic polymer that helps improve the behavior of therapeutic particles in the body. The PEG layer forms a hydrated, sterically protective surface that can reduce protein adsorption and recognition by the mononuclear phagocyte system, thereby increasing colloidal stability and potentially extending circulation time. In medicine, this platform can support the transport of poorly soluble drugs, nucleic acids, or other therapeutics, while nanoparticle size, surface properties, and drug-release design influence biodistribution and cellular uptake. These features make pegylation useful for improving pharmacokinetics, efficacy, and tolerability.

Plant viral nanoparticles (VNPs) are promising platforms for applications in biomedicine. Here, we describe the procedures for plant VNP propagation, purification, characterization, and bioconjugation. Finally, we show the application of VNPs for tumor homing and imaging using a mouse xenograft model and fluorescence imaging.

This protocol details the synthesis, hemoglobin encapsulation, and surface PEGylation of hierarchically porous UiO-66 nanoparticles designed for oxygen delivery. Type B gelatin-based engineered nanovectors system (GENS) was developed for systemic gene delivery and transfection in the treatment of pancreatic cancer. By modification with epidermal growth factor receptor (EGFR) specific peptide on the surface of nanparticles, they could target on EGFR receptor and release plasmid under reducing environment, such as high intracellular glutathione concentrations.
