Peptide Permeability

Peptide permeability is the ability of peptide molecules to cross biological barriers such as cell membranes and epithelial layers, a key property in evaluating peptide-based medicines. Permeation depends on factors including molecular size, charge, hydrophobicity, peptide sequence, membrane composition, and available transport pathways; peptides may cross through passive diffusion, transporter-mediated uptake, or endocytosis. Measuring and optimizing these processes helps researchers assess oral, transdermal, and intracellular delivery, where poor membrane penetration can limit therapeutic effectiveness. Permeability studies therefore guide peptide design, formulation, and the development of delivery systems that improve tissue access while preserving biological activity.

An Assay for Permeability of the Zebrafish Embryonic Neuroepithelium

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.
