Rde Treated Serum

RDE-treated serum is a serum sample incubated with receptor-destroying enzyme (RDE) to reduce nonspecific substances that can interfere with antibody testing, particularly in influenza serology. RDE enzymatically removes receptor-like components, including sialic acid-containing molecules that may bind virus or red blood cells independently of specific antibodies; laboratories then typically heat-treat and dilute the sample before analysis. This preparation improves the specificity and interpretability of hemagglutination-inhibition assays, which measure whether antibodies prevent influenza virus from agglutinating red blood cells. By limiting nonspecific inhibition, RDE treatment supports more reliable assessment of antibody responses in clinical studies, surveillance, and vaccine research.

The Hemagglutination Inhibition Assay to Detect Serum Antibodies Against a Target Antigen

2025

The video demonstrates a hemagglutination inhibition assay for detecting serum antibodies against specific antigens. Confirmation of hemagglutination inhibition in the test well and hemagglutination in the control well is achieved through distinct visual patterns. This assay plays a crucial role in assessing immune response, vaccine effectiveness, and studying viral infections in virology and immunology.

This protocol describes a method for extracting small RNAs from human serum. We have used this method to isolate microRNAs from cancer serum for use in DNA arrays and also singleplex quantitative PCR. The protocol utilizes phenol and guanidinium thiocyanate reagents with modifications to yield high quality RNA.
