Stem Cell Engraftment

Stem cell engraftment is the process by which transplanted stem cells migrate to a recipient’s tissues, establish themselves, and begin producing functional cell populations, making it essential to successful regenerative and hematopoietic therapies. In hematopoietic stem cell transplantation, infused cells home to bone marrow niches, adhere to supportive microenvironments, and proliferate to restore blood-cell formation and immune function after conditioning treatment. Clinicians assess engraftment through measures such as neutrophil and platelet recovery, while researchers study cell dose, tissue compatibility, immune responses, and donor chimerism to improve outcomes. Understanding this process also supports advances in gene therapy, tissue repair, and personalized medicine.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
