Vascular Pathology Study

Vascular pathology study examines structural and functional abnormalities in blood vessels and their effects on tissue perfusion, making it essential for understanding many cardiovascular and systemic diseases. It links changes such as endothelial injury, inflammation, plaque formation, thrombosis, vessel wall weakening, or abnormal dilation to altered blood flow and ischemic damage. In medicine, vascular pathology supports the diagnosis and classification of atherosclerosis, vasculitis, aneurysms, and other vascular disorders through clinical assessment, imaging, laboratory analysis, and microscopic examination. Its findings guide risk assessment, treatment decisions, and research into therapies that protect vessel integrity and improve circulation.

In Vitro Model of Physiological and Pathological Blood Flow with Application to Investigations of Vascular Cell Remodeling

This protocol replicates physiological or pathological blood flow in vitro to aid in determining cell response in disease pathologies. By introducing a pressure damping chamber downstream of a blood pump, blood flow across the vasculature can be recapitulated and imposed on a monolayer of vascular endothelium or a mimetic co-culture.

We describe a microsurgical approach for the generation of an arteriovenous (AV) loop as a model for analyzing vascularization in vivo in an isolated and well-characterized environment. This model is not only useful for the investigation of angiogenesis, but is also optimally suited for engineering axially vascularized and transplantable tissues.
