Cortical Hippocampal Neurons

Cortical hippocampal neurons are specialized nerve cells found in the cerebral cortex and hippocampus, regions that support perception, learning, memory, and cognition. They communicate by generating action potentials and releasing neurotransmitters at synapses; excitatory glutamatergic signaling and activity-dependent changes in synaptic strength help form and refine neural circuits. In neuroscience, cultured cortical and hippocampal neurons provide tractable models for studying neuronal development, synaptic plasticity, network activity, and mechanisms of neurological disease, while supporting evaluation of drugs and neuroprotective strategies.

Low-Density Primary Hippocampal Neuron Culture

This article describes the protocol for culturing low-density primary hippocampal neurons growing on glass coverslips inverted over a glial monolayer. The neuron and glial layers are separated by paraffin wax beads. The neurons grown by this method are suitable for high-resolution optical imaging and functional assays.
