Ganglia Isolation

Ganglia isolation is the process of separating clusters of neuronal cell bodies and supporting glial cells from surrounding nervous tissue for focused study. In neuroscience, researchers use precise dissection under a microscope to identify ganglia, remove adjoining tissue, and preserve the isolated structure or dissociate it into individual cells for analysis. The preparation supports investigations of neuronal morphology, connectivity, signaling, electrophysiology, and gene expression in a defined biological context. Isolated ganglia can also provide experimental material for cell culture, pharmacological testing, and studies of sensory, autonomic, or peripheral nervous system function.

Isolation of Cells from Sensory Ganglia

This video demonstrates the isolation of cells from the sensory ganglia of mice. Ganglia are subjected to enzymatic digestion to dissolve the extracellular matrix. Single cells are then collected via filtering through a cell strainer followed by density gradient centrifugation for debris removal. Finally, washing and centrifugation are done to collect the isolated cells for future analysis.

Hydraulic Extrusion of the Spinal Cord and Isolation of Dorsal Root Ganglia in Rodents

Here, we present a protocol for hydraulic extrusion of the spinal cord as well as identification and isolation of specific dorsal root ganglia (DRGs) in the same rodent. Compared to standard spinal cord isolation methods, this method is significantly faster and reduces the risk of tissue damage.
