Gender Specific Biomarkers

Gender-specific biomarkers are measurable biological features whose levels or patterns vary systematically among gender groups and may help characterize health, disease, or treatment response. In neuroscience, they can reflect sex-related biology, gendered experiences, or their interaction, including differences in hormones, gene expression, immune signaling, and neural activity. Researchers identify these markers by comparing profiles across groups and testing whether observed differences are associated with neurological conditions or clinical outcomes. Such biomarkers can improve patient stratification, support more precise research on neurological and psychiatric disorders, and clarify how biological and social factors shape brain health while promoting study designs that represent diverse populations.

Biomarker Identification for Gender Specificity of Alzheimer's Disease Based on the Glial Transcriptome Profiles

This study analyzed single-nuclei transcriptomes of thirty-three individuals with Alzheimer's disease (AD), revealing sex-specific DEGs in glial cells. Functional enrichment analysis highlighted synaptic, neural, and hormone-related pathways. Key genes, namely NLGN4Y and its regulators, were identified, and potential therapeutic candidates for gender-specific AD were proposed.

The protocol describes the development of two rodent models mimicking gender-affirming hormone therapies through subcutaneous administration of testosterone or estradiol plus cyproterone acetate (used in human therapies for transgender people): setting of the doses, identification of relevant biomarkers, and evaluation of the effects.

Skeletal Muscle Gender Dimorphism from Proteomics

A straight-forward set of methods to isolate and determine the identity of the most abundant proteins expressed in skeletal muscle. About 800 spots are discerned on a two-dimensional gel from 10 mg muscle; this allows for the determination of gender-specific protein expression. These methods will give equivalent results in most tissues.

We describe a rat model of post traumatic stress disorder (PTSD) that reveals the persistent alterations in neuroendocrine function and the delayed long-term, exaggerated fear response, characteristic of PTSD patients. The animal model and methods described here are useful for correlating biomarkers in brain nuclei, which are mechanistic but cannot be measured in patients, with biomarkers in peripheral white blood cells, which can.
