Region-specific Protein Analysis

Region-specific protein analysis is the measurement and comparison of protein abundance, localization, or molecular state within defined anatomical areas, making it possible to link protein changes to neural circuits and brain function. In neuroscience, researchers isolate a selected region or cell population, extract its proteins, and identify or quantify them using approaches such as immunoassays, electrophoresis, or mass spectrometry; comparisons across regions, conditions, or time points reveal spatial patterns. This analysis helps characterize signaling pathways, synaptic proteins, and disease-associated changes while preserving anatomical context. It can support studies of development, behavior, neurodegeneration, and responses to therapeutic intervention.

Micro-dissection of rat brain into various regions is extremely important for the study of different neurodegenerative diseases. This video demonstrates micro-dissection of four major brain regions include olfactory bulb, frontal cortex, striatum and hippocampus in fresh rat brain tissue. Useful tips for quick removal of respective regions to avoid RNA and protein degradation of the tissue are given.

This protocol presents a method to perform quantitative, single-cell in situ analysis of protein expression to study lineage specification in mouse preimplantation embryos. The procedures necessary for collection of blastocysts, whole-mount immunofluorescent detection of proteins, imaging of samples on a confocal microscope, and nuclear segmentation and image analysis are described.
