Serial Passages Colony Formation

Serial passages colony formation is a cell-culture method used to evaluate whether cells can repeatedly proliferate and generate distinct colonies under controlled conditions. During serial passage, cells are transferred from one culture to fresh conditions, and colony formation is assessed across successive rounds; sustained colony production indicates continued proliferative or self-renewal capacity. In neuroscience, this approach helps characterize neural stem and progenitor cell populations, compare growth behavior, and monitor how experimental conditions affect their maintenance or differentiation potential. Results can support studies of neural development, disease modeling, and cell-based research by linking culture performance with the persistence of relevant cellular properties.

Keratinocyte Colony Formation Assay: An In Vitro Assay to Assess the Ability of Keratinocytes to Grow into Colonies

2023

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

Passaging Cells

Cell lines are frequently used in biomedical experiments, as they allow rapid culture and expansion of cell types for experimental analysis. Cell lines are cultured under similar conditions when compared to freshly-isolated, or primary, cells, but with some basic important differences: (i) cell lines require their own specific growth factor cocktails and (ii) their growth must be more closely monitored than primary cells, as the mutations that allow them to be grown indefinitely also can...

Colony Formation Assay: Assessing the Efficacy of Anticancer Agents on Colony-Forming Lung Cancer Cells

2023

This video describes the colony formation assay, which helps determine lung cancer cells' ability to proliferate indefinitely and form individual colonies. We can analyze the effect of an anticancer agent on the colony-forming cells by analyzing the changes in the number and/or size of the resulting colonies.

2012

We provide a simple, semi-quantitative method to investigate biofilm formation in vitro. This method takes advantage of the Zeiss stemi 2000-C Dissecting Microscope (with camera attachment) to monitor both the timing and pattern of biofilm formation, as assessed by the development of wrinkled colonies.

The soft agar colony formation assay is a method used to confirm cellular anchorage-independent growth in vitro. The goal of this protocol is to illustrate a stringent method for the detection of the tumorigenic potential of transformed cells and the tumor suppressive effects of proteins on transformed cells.
