Spontaneous Coiling Activity

Spontaneous coiling activity is the rhythmic, self-generated bending and unbending of an embryo, reflecting early nervous system function before mature voluntary movement develops. In developing neural circuits, transient electrical activity and interactions among motor neurons, interneurons, and muscles produce alternating contractions that can occur without external stimulation. Studying these movements provides a visible measure of early sensorimotor development and helps researchers examine how neural networks assemble, coordinate muscle activity, and establish locomotor patterns. In neuroscience, spontaneous coiling is therefore used to assess developmental abnormalities, compare genetic or environmental conditions, and evaluate how early activity influences later motor function.

Spontaneity

A spontaneous process is one that occurs naturally under certain conditions. A nonspontaneous process, on the other hand, will not take place unless it is “driven” by the continual input of energy from an external source. Processes have a natural tendency to occur in one direction under a given set of conditions. Water will naturally flow downhill (spontaneous process), but uphill flow (nonspontaneous process) requires outside intervention such as the use of a pump. Iron exposed to the earth’s...

We describe a framework incorporating straightforward biochemical and computational analysis to guide the characterization and crystallization of large coiled-coil domains. This framework can be adapted for globular proteins or extended to incorporate a variety of high-throughput techniques.

Acute Mouse Brain Slicing to Investigate Spontaneous Hippocampal Network Activity

This protocol describes the preparation of horizontal hippocampal-entorhinal cortex (HEC) slices from mice exhibiting spontaneous sharp-wave ripple activity. Slices are incubated in a simplified interface holding chamber and recordings are performed under submerged conditions with fast-flowing artificial cerebrospinal fluid to promote tissue oxygenation and the spontaneous emergence of network-level activity.
