Flecainide Propafenone

Flecainide and propafenone are Class Ic antiarrhythmic drugs that alter cardiac electrical conduction to help control abnormal heart rhythms. They block fast sodium channels, reducing the phase 0 upstroke of cardiac action potentials and slowing impulse conduction; propafenone also has beta-adrenergic blocking activity. Clinicians use these agents to restore or maintain sinus rhythm in selected patients with atrial fibrillation, atrial flutter, and other supraventricular tachyarrhythmias. Because their strong conduction effects can widen the QRS complex and provoke serious ventricular arrhythmias, particularly in patients with structural heart disease, ECG monitoring and careful patient selection are essential in pharmacology and clinical practice.

A Fluorescent Screening Assay for Identifying Modulators of GIRK Channels

A real-time screening procedure for identifying drugs that interact with G protein-gated inward rectifier K+ (GIRK) channels is described. The assay utilizes membrane potential-sensitive fluorescent dyes to measure GIRK channel activity. This technique is adaptable for use on a number of cell lines.

Basophil activation test is a potent tool for the detection of IgE-dependent allergies in vitro. Here, an optimized protocol for basophil activation test is used to investigate drug hypersensitivity. A method for the efficient production of covalent drug-protein conjugates and their physicochemical characterization is described.
