Verapamil

Verapamil is a non-dihydropyridine calcium channel blocker used in pharmacology to examine how modulation of calcium entry influences cardiovascular function. It inhibits L-type calcium channels in cardiac muscle, the atrioventricular node, and vascular smooth muscle, reducing myocardial contractility, slowing atrioventricular conduction, and promoting vasodilation. These effects support its clinical use in hypertension, angina, and selected supraventricular tachyarrhythmias, while also illustrating how ion-channel pharmacology can alter heart rate, conduction, and vascular resistance. Studying verapamil helps relate molecular targets to therapeutic effects, drug interactions, and adverse responses such as bradycardia or impaired cardiac conduction.

Here, we present a protocol for generating chemoresistant cells using a dose-escalation approach, followed by a CRISPR/Cas9-based screen using a focused sgRNA library targeting epigenetic modifiers to identify regulators of acquired chemoresistance. Side Population Assay: A Method to Isolate Side Population of Tumor Cells from Zebrafish

The following protocol describes a method for isolating the side population of T-cell Acute Lymphoblastic Leukemia (T-ALL) tumor cells, consisting of stem cells, from an adult Zebrafish. The side population lacks prominent cell markers and hence we use Hoechst 33342 dye efflux assay to isolate them.

Isolation of Murine Embryonic Hemogenic Endothelial Cells

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
