This article details the process of isoelectric focusing (IEF) using a pre-assembled IEF cell to separate components of a plant extract based on their isoelectric points (pI). The method involves applying a high voltage to create a pH gradient, allowing for the fractionation of differently charged components.
Liquid phase isoelectric focusing enables the separation of complex bioactive molecules from crude plant extracts while maintaining their native state, supporting early-stage target validation and mechanistic de-risking in natural product discovery. This technique provides purified fractions for downstream screening, reducing biological ambiguity in hit identification workflows. By isolating components based on isoelectric point, it enhances predictive confidence in lead selection from botanical sources.
Liquid phase IEF fits within the discovery continuum from early natural product screening through hit-to-lead optimization, particularly when charge-based separation is required to resolve complex mixtures.
