This study investigates the effects of pre-formed fibrils (PFFs) on primary embryonic mouse dopamine neurons. The methodology involves quantifying the presence of phosphorylated alpha-synuclein aggregates in these neurons using fluorescence imaging techniques.
This method enables quantitative assessment of alpha-synuclein aggregation in primary embryonic mouse dopamine neurons, providing a disease-relevant system for evaluating target engagement and mechanistic de-risking in neurodegenerative disease programs. By measuring phosphorylated alpha-synuclein-positive cells, it supports predictive confidence in early discovery and informs go/no-go decisions for therapeutic candidates targeting protein misfolding pathways. The assay format in multi-well plates facilitates scalability and reproducibility for hit validation and lead optimization workflows.
The assay fits within the discovery continuum from target validation through lead identification to preclinical efficacy testing, offering a quantifiable, mechanism-based readout for alpha-synuclein-targeted therapeutics.
