This article describes a method for targeted delivery of substances to the brain in anesthetized mice by bypassing systemic circulation. The technique involves manipulating the common carotid artery and external carotid artery to infuse a test substance directly into the brain parenchyma.
This microsurgical infusion technique enables targeted delivery of test substances to the mouse brain via the internal carotid artery, bypassing systemic circulation to enhance compound bioavailability and reduce off-target effects. The method supports mechanistic de-risking in early neuroscience drug discovery by allowing precise pharmacokinetic and pharmacodynamic evaluation of CNS-penetrant molecules. It provides a reproducible platform for evaluating blood-brain barrier penetration and brain parenchyma exposure, informing go/no-go decisions in lead optimization.
The technique fits within the discovery continuum from early target validation through lead identification to preclinical efficacy testing, where controlled brain delivery is essential for evaluating CNS activity.
