Her beskriver vi fremstilling og anvendelse af et aktivitetsbaseret sonde (ARN14686, undec-10-ynyl- N – [(3 S) -2-oxoazetidin-3-yl] carbamat), der giver mulighed for påvisning og kvantificering af aktive form af det proinflammatoriske enzym N -acylethanolamine syre amidase (NAAA), både in vitro og ex vivo.
Aktivitetsbaseret proteinprofilering (ABPP) er en metode til identifikation af et enzym af interesse i en kompleks proteom ved anvendelse af en kemisk probe, som er målrettet mod enzymets aktive steder. En reporter tag indført i proben muliggør påvisning af det mærkede enzym ved in-gel fluorescens scanning, protein-blot, fluorescensmikroskopi eller væskekromatografi-massespektrometri. Her beskriver vi fremstilling og anvendelse af forbindelsen ARN14686, et klik kemi aktivitetsbaseret sonde (CC-ABP), som selektivt genkender enzymet N -acylethanolamine syre amidase (NAAA). NAAA er en cystein hydrolase, der fremmer inflammation ved at deaktivere endogene peroxisomproliferatoraktiveret receptor (PPAR) -alpha agonister, såsom palmitoylethanolamid (PEA) og oleoylethanolamide (OAS). NAAA syntetiseres som en inaktiv fuld længde proenzym, som aktiveres af autoproteolysis i det sure pH i lysosomer. Lokalisering undersøgelser have vist, at NAAA overvejende udtrykkes i makrofager og andre monocyt-afledte celler, såvel som i B-lymfocytter. Vi giver eksempler på, hvordan ARN14686 kan anvendes til at påvise og kvantificere aktiv NAAA ex vivo i gnavere væv ved protein-blot og fluorescensmikroskopi.
Almindeligt anvendte metoder til at undersøge ekspressionsmønstre, interaktioner og funktioner af proteiner, herunder væskekromatografi-massespektrometri platforme til shotgun analyse 1,2, gær to-hybrid metoder 3,4, og in vitro-assays, er begrænset ved, at de er ude af stand til at vurdere aktiviteten af proteiner i deres native tilstand. Aktivitetsbaseret protein profilering (ABPP) kan bruges til at udfylde dette hul. I denne fremgangsmåde, små-molekyle prober i stand til kovalent binding til det aktive sted af et enzym af interesse er konjugeret til en reportergruppe, der giver mulighed for måldetektion. Brug af klik-kemi (CC), kan reporter integreres i proben eller kan indføres efter mål indgreb har fundet sted 5,6. Sidstnævnte procedure kræver brug af sonder, der indeholder relevante kemiske grupper, såsom en terminal alkyn eller azid, hvilket kan modificeres med en række reporter reagenser via bio-ortogonale reaktioner such som Cu (I) -catalyzed Huisgen [3 + 2] cycloaddition 7-9 eller Staudinger ligering 10,11.
For nylig beskrev vi forbindelsen ARN14686 som den første ABP til in vitro og in vivo detektion af cystein hydrolase, NAAA 12. NAAA katalyserer den hydrolytiske deaktivering af mættede og enkeltumættede Faes, herunder oleoylethanolamide (OAS) og palmitoylethanolamid (PEA), som er endogene agonister af den antiinflammatoriske nuklear receptor PPAR-alpha 13-15. NAAA udtrykkes overvejende i makrofager og andre monocyt-afledte celler, såvel som i B-lymfocytter 14,16, hvilket antyder en rolle i reguleringen af den medfødte immunreaktion. Enzymet syntetiseres i ru endoplasmatisk reticulum i en inaktiv form og aktiveres i sure rum i cellen med en autoproteolytisk mekanisme 17. Den autoproteolytisk spaltning genererer en ny N-terminal cystein (C131 i mus og rotter, C126 i mennesker), der er nukleofilen ansvarlig for FAE hydrolyse 18,19. Farmakologisk hæmning af NAAA aktivitet ændrer FAE syntese / nedbrydning balance til fordel for øget cellulære niveauer af FAES 16,20,21. Adskillige β-lacton og β-lactam-derivater har vist sig at inhibere NAAA aktivitet med høj styrke og selektivitet 16,22-26. Disse inhibitorer virker gennem S-acylering af den katalytiske cystein 16,27,28.
Forbindelsen ARN14686 designet baseret på den kemiske struktur af den systemisk aktive, serin-afledt β-lactam NAAA inhibitor, ARN726 (4-cyclohexylbutyl- N – [(S) -2-oxoazetidin-3-yl] carbamat) 16. Det 4-butyl-cyclohexylgruppe af ARN726 blev erstattet med en C9 mættet alifatisk kæde, der bærer en terminal alkyn tag for efterfølgende CC konjugering med et azid-bærende reporter tag. Vi valgte at designe en to-trins ABP at minimally ændre strukturen af det oprindelige stillads opretholder således affiniteten af probe for NAAA. Desuden undgår indførelsen af voluminøse tags, kan en sådan probe være mere egnet til in vivo-behandling end en direkte ABP. ARN14686 inhiberer NAAA med stor styrke (hNAAA IC50 = 6 nM, rNAAA IC50 = 13 nM) ved at danne en kovalent addukt med den katalytiske cystein af enzymet 12. Forsøg i levende rotter viste, at sonden er selektiv i at fange NAAA udtrykt i lungerne. Acid ceramidase, andet cystein amidase der deler 33-34% identitet med NAAA, blev også identificeret som en lav-affinitet mål, når ved hjælp af høj probe koncentrationer (10 uM in vitro, 10 mg / ml intravenøs, iv) 12. Vi har også benyttet ARN14686 at undersøge tilstedeværelsen af aktive NAAA i betændte rottevæv efter indgivelse af komplet Freunds adjuvans (CFA) 29.
Her vil vi skitsere en protokol for preparatipå af ARN14686 (figur 1) og dens anvendelse på undersøgelsen af NAAA aktivering ex vivo. Som et eksempel beskriver vi en eksperimentel procedure at visualisere NAAA i rotte poter efter CFA administration. I dette forsøg er proteiner ekstraheret fra paw væv efter intravenøs injektion af sonden, og ABP-mærkede proteom underkastes CC med biotin-azid. Biotinylerede prøver er beriget ved hjælp streptavidinkugler, og protein blots udføres. I en anden anvendelse, beskriver vi lokalisering af aktiv NAAA ved fluorescensmikroskopi i muselunger fra probe-behandlede mus. I dette tilfælde er vævet i snit og sektioner underkastes CC for rhodamin tilsætning. En arbejdsgang skema er illustreret i figur 2.
Enzymaktiviteten fint reguleret på forskellige niveauer, herunder RNA-transkription, proteinsyntese, protein translokation, post-translationel modifikation, og protein-protein-interaktion. Ofte enzym udtryk alene tager ikke højde for dets aktivitet. ABPP blev udviklet til at studere aktiviteten af proteiner i deres native tilstand. To funktioner er nødvendige: en kemisk probe, som kovalent binder til det aktive sted af et enzym af interesse og et reporter-tag til påvisning af proben-mærket enzym.
