Peptide Quantitation

Peptide quantitation is the measurement of peptide amount or concentration in a sample, an essential step in biochemistry for evaluating synthesis, purification, and experimental dosing. It relies on signals linked to peptide content, such as ultraviolet absorbance from peptide bonds or aromatic residues and color formation in assays based on peptide-driven copper reduction; results can vary with sequence and composition. Researchers use peptide quantitation to compare purification fractions, assess recovery, normalize samples for enzymatic or binding assays, and evaluate synthetic peptide yield. Appropriate standards and complementary methods improve concentration estimates and support reproducible biochemical research.

Biochemical assays with recombinant human MHC II molecules can provide rapid, quantitative insights into immunogenic epitope identification, deletion, or design. Here, a peptide-MHC II binding assay scaled to 384-well plates is described. This cost effective format should prove useful in the fields of protein deimmunization and vaccine design and development.

Species Determination and Quantitation in Mixtures Using MRM Mass Spectrometry of Peptides Applied to Meat Authentication

We present a protocol for identifying and quantifying the components in mixtures of species possessing similar proteins. Mass spectrometry detects peptides for identification, and gives relative quantitation by ratios of peak areas. As a tool food for fraud detection, the method can detect 1% horse in beef.
