Buffy Coat Isolation

Buffy coat isolation is a laboratory method for separating leukocytes and platelets from whole blood, producing a concentrated layer enriched in nucleated cells between plasma and erythrocytes. During density-based centrifugation, red blood cells sediment because they are denser, while plasma remains above them and the intermediate buffy coat can be carefully collected. In bioengineering, this fraction provides accessible primary cells for immune-cell studies, biomaterial evaluation, tissue engineering, and blood-derived therapeutic research. Its use supports cell characterization, downstream culture, and analysis of cell-material interactions, while standardized handling helps preserve sample quality and improve reproducibility.
