Cell-derived Matrix

Cell-derived matrix is an extracellular matrix produced and deposited by living cells, providing structural support and biochemical signals that influence cell behavior. In bioengineering, researchers culture cells under controlled conditions so they assemble matrix components such as collagen, fibronectin, and proteoglycans, then may remove the cells while preserving the matrix architecture and signaling cues. These materials can serve as biologically active scaffolds for tissue engineering, cell culture, wound repair, and regenerative medicine. By reproducing aspects of the native cellular microenvironment, cell-derived matrix helps regulate adhesion, migration, proliferation, and differentiation, supporting the development of more physiologically relevant engineered tissues.

Integration Free Derivation of Human Induced Pluripotent Stem Cells Using Laminin 521 Matrix

Robust derivation of human induced pluripotent stem (hiPS) cells was achieved by using non-integrating Sendai virus (SeV) vector mediated reprogramming of dermal fibroblasts. hiPS cell maintenance and clonal expansion was performed using xeno-free and chemically defined culture conditions with recombinant human laminin 521 (LN-521) matrix and Essential E8 (E8) Medium.

The extracellular matrix plays a major role in defining the microenvironment of cells and in modulating cell behavior and phenotype. We describe a rapid method for the isolation of cell-derived extracellular matrix, which can be adapted to different scales for microscopic, biochemical, proteomic, or functional studies.

The tissue-specific extracellular matrix (ECM) is a key mediator of cell function. This article describes methods for synthesizing pure ECM-derived foams and microcarriers that are stable in culture without the need for chemical crosslinking for applications in advanced 3D in vitro cell culture models or as pro-regenerative bioscaffolds.

Fabricating a Kidney Cortex Extracellular Matrix-Derived Hydrogel

Here we present a protocol to fabricate a kidney cortex extracellular matrix-derived hydrogel to retain the native kidney extracellular matrix (ECM) structural and biochemical composition. The fabrication process and its applications are described. Finally, a perspective on using this hydrogel to support kidney-specific cellular and tissue regeneration and bioengineering is discussed.

Fabrication of Decellularized Cartilage-derived Matrix Scaffolds

Decellularized cartilage-derived scaffolds can be used as a scaffold to guide cartilage repair and as a means to regenerate osteochondral tissue. This paper describes the decellularization process in detail and provides suggestions to use these scaffolds in in vitro settings.
