Cell Morphology

Cell morphology is the study and characterization of a cell’s shape, size, structure, and spatial organization, features that reflect its function and response to the surrounding environment. In bioengineering, researchers assess morphology through microscopy and image analysis, relating changes in cell spreading, alignment, adhesion, and organization to substrate properties, mechanical forces, biochemical cues, or disease-related conditions. Morphological measurements help evaluate engineered tissues, biomaterials, organ-on-chip systems, and cell-based therapies by indicating cellular responses and tissue maturation. Combined with molecular and functional assays, morphology can clarify how engineered microenvironments regulate cell behavior and guide the design of regenerative and diagnostic technologies.

We present a protocol that permits to view and to quantitatively asses the morphology of the dendritic tree of individual Purkinje cells grown in organotypic cerebellar slice cultures. This protocol is intended to promote studies on the mechanisms of Purkinje cell dendritic development.

Bone Marrow Aspiration: A Method to Obtain Bone Marrow to Examine Cell Morphology

2023

This video describes the technique of aspirating bone marrow from iliac spine and smear preparation for biopsy. In the example, we perform bone marrow aspiration from an acute myeloid leukemia (AML) patient and examine the cells to check for measurable residual disease (MRD), an important prognostic biomarker.

Whole-cell Patch-clamp Recordings from Morphologically- and Neurochemically-identified Hippocampal Interneurons

Cortical networks are controlled by a small, but diverse set of inhibitory interneurons. Functional investigation of interneurons therefore requires targeted recording and rigorous identification. Described here is a combined approach involving whole-cell recordings from single or synaptically-coupled pairs of neurons with intracellular labeling, post-hoc morphological and immunocytochemical analysis.

We have developed a computer program to analyze neuronal morphology. In combination with two existing open source analysis tools, our program performs Sholl analysis and determines the number of neurites, branch points, and neurite tips. The analyses are performed so that local changes in neurite morphology can be observed.
