Collagen Substrate Preparation

Collagen substrate preparation is the process of producing collagen-coated surfaces or collagen-based matrices that provide cells with a biologically relevant material for attachment and growth. It typically involves dissolving or diluting collagen, adjusting pH and concentration, applying the material to a culture surface, and allowing it to adsorb or polymerize under controlled temperature conditions. The resulting substrate presents extracellular-matrix cues that can influence cell adhesion, spreading, migration, and phenotype. In bioengineering, prepared collagen substrates support in vitro cell culture, tissue-engineering studies, biomaterial evaluation, and the development of models for investigating cell–matrix interactions.

Traditional procedures for the isolation of soluble type 1 collagen (COL1) require about 10 days from start to finish because of lengthy buffer incubations and laborious resuspensions of fibrils. Here, we describe a means to purify COL1 from small dermal biopsies in less than 3 hr.

Evaluation of Keratinocyte Proliferation on Two- and Three-dimensional Type I Collagen Substrates

Here, we present a protocol for the preparation of two different forms of culture substrates utilizing type I collagen. Depending on how collagen is handled, collagen molecules either maintain two-dimensional, non-fibrous form or reassemble into three-dimensional, fibril form. Cell proliferation on type I collagen is drastically affected by fibril formation.
