Lower Limb Amputation

Lower limb amputation is the surgical removal of part or all of a lower extremity, a life-changing intervention used to remove nonviable tissue or manage severe injury and disease while preserving function where possible. It involves dividing bone and soft tissues, shaping and protecting the residual limb, and allowing healing before rehabilitation and prosthetic fitting. In bioengineering, knowledge of anatomy, biomechanics, tissue loading, and neural control guides the design of prosthetic limbs, sockets, sensors, and powered devices that support stable, energy-efficient movement. These technologies can improve mobility and independence, while research on implantable interfaces and personalized control aims to reduce discomfort, enhance sensory feedback, and enable more natural gait.

Lower limb amputation may occur even after angioplasty of obstructed vessels in Critical Limb Ischemia (CLI). Mononuclear Progenitor Cells (MPCs) reflect vascular repair. The present protocol describes the quantification of MPCs from circulation close to angioplasty, and its relationship with endothelial dysfunction and prediction of lower limb amputation.
