Resistance Artery Mechanics

Resistance artery mechanics describes how small arteries and arterioles deform, generate force, and regulate blood flow in response to pressure and vascular signals. Their walls combine elastic components with smooth muscle, whose contraction or relaxation changes vessel diameter, wall tension, and hydraulic resistance, thereby controlling tissue perfusion and influencing blood pressure. In bioengineering, studying these mechanics supports the design of vascular models, microfluidic systems, and biomaterials that reproduce physiological flow conditions. Mechanical measurements and computational analyses also help clarify how altered stiffness, remodeling, or contractility contributes to vascular dysfunction and guide the development of diagnostic and therapeutic strategies.

In pressure myography, an intact small segment of a vessel is mounted onto two small cannulas and pressurized to a suitable luminal pressure. Here, we describe the method to measure vasorelaxation response of the mouse 3rd order mesenteric arteries in c57 and sGCα1-/- mice using pressure myography.

A cell culture model of resistance arteries is described, allowing for the dissection of signaling pathways in endothelium, smooth muscle, or between endothelium and smooth muscle (the myoendothelial junction). The selective application of agonists or protein isolation, electron microscopy, or immunofluorescence can be utilized using this cell culture model.
