3d Cell Imaging

3D cell imaging is a group of microscopy techniques that captures and reconstructs the three-dimensional structure of cells, revealing spatial relationships that conventional two-dimensional images can miss. These methods acquire optical sections at different depths, often using fluorescent labels with confocal, multiphoton, or light-sheet illumination, then combine the sections into a volumetric model. In biology, 3D cell imaging helps researchers examine organelle organization, cell morphology, migration, division, and interactions within tissues or organoids. Quantitative analysis of these datasets can clarify cellular mechanisms, support disease research, and improve evaluation of treatments in physiologically relevant models.

Rapid Acquisition of 3D Images Using High-resolution Episcopic Microscopy

We describe a detailed protocol using high-resolution episcopic microscopy to acquire three-dimensional (3D) images of mouse embryos. This improved protocol utilizes a modified tissue preparation method to enhance penetration of the fluorescent dye, thereby permitting morphometric analysis of both small and large-sized specimens.
