Primary Preadipocytes

Primary preadipocytes are progenitor cells isolated directly from adipose tissue that can develop into mature fat-storing adipocytes, making them valuable models of adipose biology. Under appropriate culture conditions, adipogenic signals activate transcriptional regulators such as PPARγ and C/EBPα, which drive coordinated changes in gene expression, cell morphology, and intracellular lipid accumulation. These cells help researchers examine adipocyte differentiation, lipid metabolism, obesity-related dysfunction, and interactions between adipose tissue and other cell types. Because they retain characteristics of the donor tissue more closely than many immortalized lines, primary preadipocytes support physiologically relevant studies of development, disease mechanisms, and potential metabolic therapies.

Isolation and Differentiation of Primary White and Brown Preadipocytes from Newborn Mice

This report describes a protocol for the simultaneous isolation of primary brown and white preadipocytes from newborn mice. Isolated cells can be grown in culture and induced to differentiate into fully mature white and brown adipocytes. The method enables genetic, molecular, and functional characterization of primary fat cells in culture.

Isolation of Preadipocytes from Broiler Chick Embryos

The present protocol describes a simple method for isolating preadipocytes from adipose tissue in broiler embryos. This method enables isolation with high yield, primary culture, and adipogenic differentiation of preadipocytes. Oil Red O staining and lipid/DNA stain measured the adipogenic ability of isolated cells induced with differentiation media.
