Adenoviral Gene Expression

Adenoviral gene expression is the process by which adenovirus genetic information is transcribed and translated in infected cells, making it important for understanding viral biology and gene delivery. After receptor-mediated entry and endosomal escape, the viral genome reaches the nucleus, where host RNA polymerase transcribes early genes that regulate the cell and viral replication, followed by late genes encoding structural proteins. In recombinant, replication-defective adenoviral vectors, these viral functions are modified to express an inserted gene rather than produce infectious virus. This system supports transient, high-level expression for studying gene function, developing vaccines, and testing gene-based therapies.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.

These studies report on reversible attachment of adenoviral gene vectors to coatless metal surfaces of stents and model mesh disks. Sustained release of transduction-competent viral particles contingent upon hydrolysis of cross-linkers used for vector immobilization results in a durable site-specific transgene expression in vascular cells and in stented arteries.

Neo-Islet Formation in Liver of Diabetic Mice by Helper-dependent Adenoviral Vector-Mediated Gene Transfer

We describe hepatic neo-islet formation in STZ (streptozotocin)-induced diabetic mice by gene transfer of Neurogenin3 (Ngn3) and Betacellulin (Btc) using helper-dependent adenoviral vector (HDAd) and the reversal of hyperglycemia. Our method takes advantages of helper-dependent adenoviral vectors with their highly efficient in vivo transduction and the long lasting gene expression.
